Retatrutide ships as a lyophilised (freeze-dried) cake and has to be dissolved in bacteriostatic water before it can be measured or used in an assay. Step-by-step reconstitution

Calculate the water volume
Decide the concentration you want to work at and divide the vial mass by it. A 10mg vial at 5 mg/mL needs 2 mL of bacteriostatic water. The volume changes concentration only — the vial always holds the same total peptide.

Bring both vials to room temperature
Take the lyophilised retatrutide vial and the bacteriostatic water out of refrigeration and let them equilibrate for 20–30 minutes. Cold solvent dissolves the cake more slowly.

Sanitise the stoppers
Wipe the rubber stopper of both the peptide vial and the bacteriostatic water vial with a fresh alcohol prep pad and let them air-dry before any needle enters.

Draw the diluent
Using a sterile syringe, draw the calculated volume of bacteriostatic water. Check the barrel for air and expel it before moving to the peptide vial.

Add the water down the vial wall
Angle the needle so the stream runs down the inside glass wall of the vial rather than striking the lyophilised cake directly. Release the plunger slowly and let the vacuum draw the last of the volume in.

Swirl — never shake
Roll or swirl the vial gently between the fingers, or invert it slowly, until the cake fully dissolves. Shaking creates foam and shear stress that can aggregate the peptide.

Inspect the solution
Hold the vial against a light source. The solution should be clear and colourless with no particulates, fibres or cloudiness. Discard the vial if anything is visible.

Label and refrigerate
Mark the vial with the compound, concentration, reconstitution date and batch number, then store at 2–8 °C protected from light. Treat the in-use window as roughly 30 days.
What the solution should look like
Correctly reconstituted retatrutide is clear and colourless, with no visible particulates. The lyophilised cake typically dissolves within a minute or two of gentle swirling; larger vials at higher concentration can take a little longer. Some vials show fine bubbles immediately after mixing, which settle on standing and are not a problem.
Discard the vial rather than using it if you see persistent cloudiness, a yellow or brown tint, fibres, flakes, or anything that will not go into solution after several minutes. Those are signs of degradation, contamination or a compromised stopper seal, and no amount of further mixing corrects them.

- Avoid freezing the solution. Freeze–thaw cycles stress the peptide. Unreconstituted lyophilised powder is what should be frozen for long-term holding.

Mistakes that ruin a vial
- Shaking. The fastest way to denature and foam a peptide. Swirl or invert slowly instead.
- Firing the water directly at the cake. Run it down the glass wall so it reaches the powder gently.
- Mixing a cold vial straight from the fridge. Let both vials reach room temperature first.
- Using tap, distilled or plain sterile water for a multi-entry vial. Only bacteriostatic water is preserved for repeated withdrawals.
- Leaving the vial on the bench. Get it back to 2–8 °C once mixed.
